Molecular biology: Blotting / Hybridization techniques
نویسنده
چکیده
What is blotting / hybridization? Blotting of nucleic acid is the central technique for hybridization studies. Double stranded DNA will denature or separate at high temperatures into single strands. When the temperature is lowered, the two strands will anneal because of the base pairing interaction between the complementary strands. If nucleotide sequences are identical or similar, complementary strands from different sources will anneal. This is called hybridization to indicate that each strand of DNA came from a different source.

Molecular Hybridization Techniques of Nucleic Acids
The nucleic acid hybridization is the process wherein two DNA or RNA single chains (mono-stranded) from different biological sources, make the double catenary configuration, based on contingent sequence homology of the two sources, resulting DNA-DNA, RNA-RNA or DNA-RNA hybrids. The purpose is identification or localization of certain nucleic acid sequences (genes) in the genome of some species....
متن کاملSouthern Blotting and Related DNA Detection Techniques
Southern blotting is one of the central techniques in molecular biology. First devised by E.M. Southern (1975), Southern blotting results in transfer of DNA molecules, usually restriction fragments, from an electrophoresis gel to a nitrocellulose or nylon sheet (referred to as a ‘membrane’), in such a way that the DNA banding pattern present in the gel is reproduced on the membrane. During tran...
متن کاملDot and slot blotting of DNA.
Dot and slot blotting are simple techniques for immobilizing bulk unfractionated DNA on a nitrocellulose or nylon membrane. Hybridization analysis can then be carried out to determine the relative abundance of target sequences in the blotted DNA preparations. Dot and slot blots differ only in the geometry of the blot, a series of spots giving a hybridization pattern that is amenable to analysis...
متن کاملNorthern blotting analysis.
Northern blotting analysis is a classical method for analysis of the size and steady-state level of a specific RNA in a complex sample. In short, the RNA is size-fractionated by gel electrophoresis and transferred by blotting onto a membrane to which the RNA is covalently bound. Then, the membrane is analysed by hybridization to one or more specific probes that are labelled for subsequent detec...
متن کاملContinuity and change: paradigm shifts in neural induction.
The problem of "primary embryonic induction" was one of the first areas of developmental biology to become "molecularized." What had been seen as an intractable series of problems became amenable to the techniques of Northern blotting, ectopic RNA insertion, and in situ hybridization. These molecular analyses showed that some of the fundamental concepts of primary embryonic induction concluded ...
